In non-apoptotic cells, the JC-1 dye enters the mitochondria, which causes a fluorescence redshift from green (~ 529 nm) to red (~ 590 nm). Depolarization of mitochondria will cause a green shift in cells, indicating that they are apoptotic.
Used for measurements using the NC-3000 for Mitochondrial Potential Analysis in combination with Solution 8.

The programmed method in the NucleoCounter® NC-3000 automatic analyzer allows the user to measure mitochondrial transmembrane potential using the cationic dye JC1 (5, 5, 6, 6-tetrachloro-1, 1, 3, 3 tetraethylbenzimidazole carbocyanine iodide).
The method describes the detection of a violation of the membrane potential of mitochondria in individual cells. It is known that the loss of mitochondrial membrane potential precedes apoptosis and necrosis caused by chemical hypoxia. The lipophilic cationic dye JC-1 exhibits potential-dependent uptake in mitochondria and is a simple fluorescent technique for distinguishing between healthy and apoptotic cells. In healthy cells, the negative charge created by the intact potential of the mitochondrial membrane promotes the accumulation of JC-1 in the mitochondrial matrix. At high concentrations, JC-1 aggregates and turns red. In apoptotic cells, the mitochondrial potential collapses and JC-1 is localized in the cytosol in its monomeric green fluorescent form.
