The S100 reagent is used to destroy the plasma membranes of sperm cells, making the nuclei susceptible to staining with propidium iodide (PI).
Used for analysis when using SP-100 ™ for total sperm count.

Description
Lysis / Dilution Buffer S100 is always added to the semen sample prior to performing a total count. Adding a buffer serves two purposes:
- Destruction of plasma membranes, making sperm nuclei sensitive to PI staining.
- Diluting the semen sample. A mixture of a semen sample and S100 reagent is called a lysate mixture or a sample / reagent mixture.
In boar ejaculates, the sperm density is usually in the range of 100 to 1400 million cells per ml. To obtain an accurate sperm count with the NucleoCounter SP-100, it is recommended to dilute the boar ejaculate 201 times (50 µl ejaculate + 10 ml S100 reagent). This corresponds to a sperm density in the range of 0.5 to 7.0 million per ml in the lysate mixture. Approximately 1 μl of the lysate mixture is counted in the SP1 cassette, which corresponds to a total of 500-7000 sperm cells per analysis.
The ejaculates of each animal species have their own optimal dilution factor, depending on the typical sperm concentration for the given species.
