Description
The Y100 reagent is used to disrupt the plasma membranes of yeast cells, making the nuclei susceptible to staining with propidium iodide (PI).
It is recommended to use a 1:10 ratio of sample volume to Y100 reagent (for example, 50 ml of sample and 450 ml of Y100 reagent).
If the sample medium is a colored solution such as beer or wine, dilution may be required before using the Y100 reagent so that the color does not affect the fluorescence intensity detected from the cells.
Used to determine the total number of yeast cells in a sample using the ChemoMetec YC-100 ™ and NC-3000 ™ instruments.
