Description

Via1-Cassette is a dedicated cell sampling and staining device for NucleoCounter instruments. The cassette contains two immobilized fluorophores, acridine orange (AO) and DAPI.

The sample is loaded into a cassette by dissolving AO and DAPI in the mixing channels. AO stains all cells in the sample, both living and dead, thus ensuring their total number; DAPI stains only dead cells.

Each cassette has an individual dot code indicating the exact volume of its counting chamber. This dot code is read by the NucleoCounter during analysis and used by the software to accurately calculate the cell concentration in each sample.

Via1-Cassette is used in Nucleocounter NC-200 and Nucleocounter NC-3000.

Loading a sample into a cassette

The cassette must be filled carefully by pressing the white piston, which creates a partial vacuum in the flow system. When the piston is pressed, the cassette tip must be immersed in the sample mixture, causing the sample to be loaded into the flow system. Approximately 60 μL is loaded into the flow system, filling the first part of the channel in the cassette.

After placing the cassette in the device, the colored mixture is automatically transferred to the measuring chamber, where the fluorescent image is recorded. The measured volume in the cassette reaches 1.4 µl.

After analysis, the sample and stains are inside the cassette, which can be safely disposed of.
Be careful when wiping the cassette as plastic material may be scratched.
