Description
The NucleoCounter® SP-100 ™ Sperm Cell Counter is easy to operate and efficiently determines the total sperm concentration and viability in a sample from ejaculates or extended doses.
The NucleoCounter SP-100 is designed as a stand-alone instrument, but if desired, it can be connected to a computer via USB or to a printer via RS-232. When connected to a computer, SemenView offers various functions such as documenting the results obtained.
The instrument is part of the NucleoCounter SP-100 System, which includes the SP1 Disposable Cassette, S100 Reagent (Lysis / Dilution Buffer), and PBS (Dilution Buffer).
The principle of operation of the system is based on DNA staining of mammalian spermatozoa using a fluorescent dye, propidium iodide (PI). When PY is added to a suspension of chemically pretreated sperm, the dye will specifically bind to the cell nucleus. If the cell suspension is subsequently illuminated with green light, the dye bound to the nuclei of the cells will emit red fluorescent light. When exposing an image using microscopic techniques, each nucleus can be seen as dots against a dark background.
The compact tool is ideal for any kind of research, quality control or production. The measuring range of the NucleoCounter® SP-100 ™ is virtually unlimited thanks to dilution prior to analysis.
NucleoCounter SP-100 is equipped with advanced imaging software. This integrated software analyzes the recorded image and counts the number of cores in the image. Since the measuring chamber volume of the disposable cassette and the dilution factor are known, the cell density in the original sample can be calculated. The sperm concentration of the original sample (expressed in millions per ml) is shown on the NucleoCounter SP-100 display.
The whole process takes approximately 30 seconds (from clicking “Run” to presenting the result). Counting non-viable cells takes about twice as long as the total count (i.e. 60 seconds) because the instrument performs more complex analysis. Cell counting with the NucleoCounter SP-100 is objective and operator independent.
